Well going via Bris, its about 3879km give or take a few.
I imagine you will want the quickest route which will be maybe out to St George and up the centre via Longreach, Mt Isa , Barkly Highway to Three ways turn right and then up the Stuart HIghway for 850km or so then you reach Darwin.
It will be hot, but nothing you can do about that other than plenty of water and eat regularly. The heat still tires you even when driving, so the key is to take it EASY. Long stretches of road do get all the same and concentration does wander. Plus you need to be alert for animals and stock etc.
You are arriving into Darwin in the buildup- poor you all if you don't like humidity :{ You may get some rain and storms closer up to the top end, so don't drive through flooded roads- sometimes the highway gets water over after alot of rain, so take it easy there.
Book a stay of maybe a few nights somewhere to chill out - could be Longreach. Yes its hot- but also dry heat and there are some good things to see.
A stop at the Bitter springs or Mataranka hot springs could be good- the caravan parks all have pools.
Car fridge a good idea. Has your car got a second battery ? Often the heat outside will heat your car up and your fridge needs to work harder to keep cold and the battery can drain faster.
Dont drive at night- have you got a bullbar or driving lights ?
You will be driving with lots of semi's and then possibly road trains so read up on remote driving and not to overtake road trains unless you have a clear 1km of vision etc.
Face it- your kids will be horribly bored and your 9mo may never want to be restrained in a car again ! lol,but they will eventually get over it.

Regretfully it may not be possible to drive only during daylight hours as there is quite a distance between towns in outback Queensland and the NT. And travelling with children you may need frequent stops to let them burn off energy.
Stops, Mitchell has artesian bores where you can paddle/swim, don't take the shorter route Morven Augahella but go into Charleville for an over night stop, there is a far better range of accommodation and meals available. Tambo a small town but visit the Tambo Teddies, the youngest would love a sheepskin Teddy Bear and they did supply them to Prince George when he was born. Longreach is worth a days stop though the children will be too young to appreciate the history on display there. Winton = dinosaurs, and some old classical outback buildings, McKinlay the pub from Crocodile Dundee and at Cloncurry there is an open air museum just near where you join the Flinders highway. Places to stay in Mt Isa can be hard to find but you can stock up on food for the next section. Three Ways has been upgraded and you may as well stay here instead of driving into Tennant Creek. At the local Tourist Information centres along the way, they will have more information about local attractions. They are staffed by knowledgeable locals.Detour into Daly Waters, it is close to the highway and also Mataranka with a day or so in Katherine.

If you are relocating to Darwin, then I would probably save the Kakadu trip for when its cooler. It will be really hot walking with your kids and really non of Kakadu can be seen from the road.
By the time you pass through Three ways you will just want to get to the end of your trip and have your own bed.
Definitely book a cabin at DW, as even though its just a pub and nothing else other than roadhouse, its can get very busy.
Same with Katherine- if you dont like the heat- yesterday it was 37 EEK. Edith Falls just out of Katherine will be worth a stop for a swim.
I would even actually skip DW and drive straight to Mataranka and stay at the choice of places there and be able to have a swim.
I would focus my time more on the QLD coast and then save your Kakadu the NT part for when you are here.
